Levels of the pheromone 7,11-heptacosadiene in the sibling species Drosophila simulans and Drosophila sechellia

This dataset provides measurements of the cuticular pheromone, 7,11-heptacosadiene of individual female vinegar flies. The purpose of the dataset is to compare the levels between two sibling species, Drosophila simulans and Drosophila sechellia. Single female virgins were isolated from males at eclosion, aged five days, and then washed in hexane to extract cuticular hydrocarbons. The extracts were analyzed by gas chromatography relative to an internal standard. Statement

Drosophila females that were five days old were anaesthetised on ice for ten minutes. Individual female flies were then transferred to 150uL pure HPLC grade hexane within a 200uL glass vial that was fitted with a glass insert. The vial with the fly inside was vortexed to extract the hydrocarbons. Afterwards the fly was removed, the hexane extract was dried down under a gentle flow of nitrogen, and the vial was shipped to Groningen for gas chromatograph analysis. The retention time in minutes of each peak relative to an internal standard (C26), was used to measure size and identify 7,11-heptacosadiene. The area underneath each peak, relative to the standard was used to measure the quantity of 7,11-heptacosadiene. ChemStation software (Agilent technologies) was used to estimate area under the peak.
