Metabolomic data from the whole-leaf and headspace of barley plants inoculated with rhizobacteria and infested with aphids

[This nonGeographicDataset is embargoed until January 4, 2027]. This dataset comprises whole-leaf and headspace metabolomics of barley plants (Hordeum vulgare) that were root-inoculated with three rhizobacterial treatments: Acidovorax radicis N35, Bacillus subtilis B171, or no bacteria (control). Plants were also either infested with Sitobion avenae aphids or left uninfested as controls. Plants were grown under controlled conditions in glasshouse (leaf extracts) or climate chamber (headspace analysis). The purpose of these experiments was to compare metabolomic profiles across rhizobacterial treatments, and when aphids were feeding on plants, as an assessment of wider-community effects on rhizosphere inoculation of barley plants. Statement

Barley plants (Hordeum vulgare, cultivars Irina (KWS seeds) and Firefoxx (Elsoms seeds)), were treated with Acidovorax radicis N35, Bacillus subtilis B171, or a control, and either infested with Sitobion avenae aphids or left uninfested. In the first experiment, whole leaves were sampled, metabolites extracted with hexane, derivatised and analysed using an Agilent 7250 GC-qToF-MS equipped with an HP-5MS column (30 m × 0.25 mm × 0.25 μm). In the second experiment, plants were enclosed in transparent PET film bags (31 cm × 30 cm; Toppits® Bratschlauch, Melitta Group, Germany) sealed at both ends and headspace volatiles were collected using Tenax probes and analysed on a GC-qToF-MS coupled with thermal desorption. Data were processed using MS-DIAL, with blank subtraction, internal standard normalisation, QC correction, and log transformation. Annotated peaks were matched against spectral libraries. Features were assigned metabolite numbers to enable unbiased comparisons, including unknowns, across both experiments.
